BMP-7 Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemical analysis of BMP-7 in immersion fixed paraffin-embedded sections of human prostate. Samples were incubated in BMP-7 monoclonal antibody (Product # MA5-23878) using a dilution of 25 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown) and counterstained with hematoxylin (blue). Lower panel shows a lack of labeling if primary antibodies are omitted and tissue is stained only with secondary antibody followed by incubation with detection reagents.

Samples were incubated in BMP-7 monoclonal antibody (Product # MA5-23878) using a dilution of 25 µg/mL overnight at 4 °C. Tissue was stained with the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown) and counterstained with hematoxylin (blue).

BMP-7 Antibody in Neutralization (Neu) — Neutralization of BMP-7 in ATDC5 mouse chondrogenic cell line. Samples were incubated in BMP-7 monoclonal antibody (Product # MA5-23878). Recombinant Human BMP-7 induces alkaline phosphatase production in the the ATDC5 mouse chondrogenic cell line in a dose-dependent manner (orange line). Alkaline phosphatase production elicited by Recombinant Human BMP-7 (1 µg/mL) is neutralized (green line) by increasing concentrations of Human BMP-7 Monoclonal Antibody. The ND50 is typically 4-12 µg/mL in the presence of L-ascorbic acid (50 µg/mL).

In ELISAs, this antibody shows approximately 25-50% cross-reactivity with recombinant human (rh) BMP-6 and no cross-reactivity with rhBMP-2, -3, -4, -5, or -8.

Reconstitute at 0.5 mg/mL in sterile PBS.

Endoxin level is <0.10 EU per 1 µg of the antibody by the LAL method.

The bone morphogenetic proteins (BMPs) are a family of secreted signaling molecules that can induce ectopic bone growth. Many BMPs are part of the transforming growth factor-beta (TGFB) superfamily. BMPs were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extra skeletal site. Based on its expression early in embryogenesis, BMP7 has a proposed role in early development. In addition, the fact that BMP7 is closely related to BMP5 and BMP6 has lead to speculation of possible bone inductive activity.
