Detection of human CDC40 by western blot and immunoprecipitation. Samples: Whole cell lysate from HeLa (15 and 50 µg for WB; 1 mg for IP, 20% of IP loaded), HEK293T (T; 50 µg) and Jurkat (J; 50 µg) cells. Antibodies: Affinity purified rabbit anti-CDC40 antibody (Product # A303-699A) used for WB at 0.1 µg/mL (A) and 1 µg/mL (B) and used for IP at 6 µg/mg lysate. Pre-mRNA splicing occurs in two sequential transesterification steps. The protein encoded by this gene is found to be essential for the catalytic step II in pre-mRNA splicing process. It is found in the spliceosome, and contains seven WD repeats, which function in protein-protein interactions. This protein has a sequence similarity to yeast Prp17 protein, which functions in two different cellular processes: pre-mRNA splicing and cell cycle progression. It suggests that this protein may play a role in cell cycle progression.
