Immunocytochemistry analysis of DHFR in HeLa cells. Nuclei were stained blue with DAPI; DHFR recombinant monoclonal antibody (Product # MA5-53398) with a dilution of 1:1,000 was stained magenta with Alexa Fluor 647. DHFR Antibody — Antibody specificity was demonstrated by siRNA mediated knockdown of target protein. HeLa cells were transfected with Dihydrofolate reductase shRNA and decrease in signal intensity was observed in Western Blotting using Anti-DHFR Recombinant Rabbit Monoclonal Antibody (23GB3575) (Product # MA5-53398) at a dilution of 1:5,000. Dihydrofolate reductase converts dihydrofolate into tetrahydrofolate, and is required for the synthesis of purines, thymidylic acid and certain amino acids. While the functional dihydrofolate reductase gene has been mapped to chromosome 5, multiple intronless processed pseudogenes or dihydrofolate reductase like genes have been identified on separate chromosomes. Dihydrofolate reductase deficiency has been linked to megaloblastic anemia.
