DLL4 Antibody (PA5-46974) in ICC/IF

Immunocytochemistry analysis of DLL4 in immersion fixed bEnd.3 mouse endothelioma cell line. Samples were incubated in DLL4 polyclonal antibody (Product # PA5-46974) using a dilution of 10 µg/mL for 3 hours at room temperature followed by NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm.

DLL4 Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemical analysis of DLL4 in immersion fixed paraffin-embedded sections of mouse embryo (13 d.p.c.). Samples were incubated with DLL4 polyclonal antibody (Product # PA5-46974) using a dilution of 5 µg/mL for 1 hour at room temperature followed by Anti-Goat IgG VisUCyte™ HRP Polymer Antibody. Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to developing vasculature.

DLL4 Antibody in Western Blot (WB) — Western blot analysis of DLL4 in bEnd.3 mouse endothelioma cell line. Samples were incubated in DLL4 polyclonal antibody (Product # PA5-46974) using a dilution of 2 µg/mL followed by a HRP-conjugated Anti-Goat IgG secondary antibody. A specific band was detected for DLL4 at approximately 90 kDa (as indicated). This experiment was conducted under reducing conditions.

DLL4 Antibody in Immunohistochemistry (IHC) — Figure 2 Distribution of M1 and M2 macrophages with Dll4 expression in laser-induced CNV observed under an immunofluorescence microscope. One eye of each C57BL/6 mouse was exposed to a laser to induce CNV, and the other eye was left untreated. Eyecups were collected on day 7 after laser coagulation. (A and B; thin white arrows) Dll4-positive M1 macrophages were marked by double staining for CD80 (green) and Dll4 (red). (C and D; thick white arrows) Dll4-positive M1 macrophages in cryosections of the CNV lesion were found expressed at low levels either untreated or after laser coagulation. Dll4-positive M2 macrophages were marked by double staining for CD206 (green) and Dll4 (red). (E) Dll4-positive M2 macrophage number in cryosections of the CNV lesion was significantly increased after laser coagulation compared with untreated eyes. Each experiment was repeated three times. Delta-like ligand 4 (DLL4), also know as Delta-4, is a homologue of the Drosophila delta protein, and functions as a transmembrane bound ligand to the notch receptor Notch1. DLL4 is expressed in the vasculature and plays a critical role in vascular development. It is induced by vascular endothelial growth factor (VEGF), as a negative feedback regulator to regulate angiogenic sprouting and promote the formulation of a differentiated vascular network. DLL4 has been found to be strongly expressed in tumor vessels of clear-cell renal tumors and bladder cancer, and inhibition of DLL4 results in increased vascular proliferation but defective maturation. This in turn leads to a decrease in tumor growth, with no apparent toxicity.
