FHOD3 is a novel member of the formin homology family playing an important role in regulation of the actin cytoskeleton. It contains a DAD (diaphanous auto regulatory) domain, an FH1 (formin homology 1) domain, an FH2 (formin homology 2) domain and a GBD/FH3 (Rho GTPase-binding/formin homology 3) domain. FHOD3 is a cytoplasmic protein where the main part of the protein localizes to actin fibers and the remaining part displays filamentous staining. A 1422 amino acid containing formin-related actin-organizing protein, FHOD3 may cause stress fiber formation together with cell elongation and may associate with nestin/NES-based interfilament (IF). Expression of FHOD3 is seen mainly in heart, kidney, and brain.
