Immunofluorescence analysis of HB9 was performed using 70% confluent log phase Caco-2 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. Homeobox protein HB9 is expressed in lymphoid and pancreatic tissues. It is reported to be Putative transcription factor involved in pancreas development and function. Defects in this gene causes Currarino syndrome. The triad of a presacral tumor, sacral agenesis and anorectal malformation constitutes the Currarino syndrome which is caused by dorsal-ventral patterning defects during embryonic development. It is also reported that HB9 is strongly expressed in epidermal basal cells of the outer scale face in tarsometatarsal scale skin.
