INSR Antibody (PA5-115426) in ICC/IF

Immunocytochemistry analysis of INSR in HepG2 cells. Samples were treated with PFA, permeabilized in 0.1% Triton X-100, blocked in 10% serum (45 min at 25°C), and incubated with polyclonal antibody (Product # PA5-115426) at a dilution of 1:200 (1 hr at 37°C). Secondary staining was applied with mouse anti-beta tubulin; AlexaFluor 594 conjugated goat anti-rabbit IgG (Red); AlexaFluor 88 conjugated goat anti-mouse IgG (Green) and DAPI (blue) using a dilution of 1:200 (1 hr at 37°C).

Secondary staining was applied with Alexa Fluor® 594 conjugated goat anti-rabbit IgG using a dilution of 1:600.

INSR Antibody in Immunocytochemistry (ICC/IF) — Immunocytochemistry analysis of INSR in MCF-7 cells. Samples were treated with PFA, permeabilized in 0.1% saponin prior to blocking in 10% serum (45 min at 37°C), and incubated with polyclonal antibody (Product # PA5-115426) at a dilution of 1:400 (1 hr at 37°C). Secondary staining was applied with Alexa Fluor® 594 conjugated goat anti-rabbit IgG using a dilution of 1:600.

INSR Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry analysis of INSR in human breast carcinoma. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. Samples were incubated with INSR polyclonal antibody (Product # PA5-115426) using a dilution of 1:100 (1.5 hours at 22°C) followed by HRP conjugated goat anti-rabbit secondary antibody.

INSR Antibody in Western Blot (WB) — Western blot analysis of INSR in SKBR-3 (left lane: treated with blocking peptide). Samples were incubated with polyclonal antibody (Product # PA5-115426).

Antibody detects endogenous levels of total IR.

靶标信息

INSR is a receptor tyrosine kinase. Binding of insulin to the extracellular domain of INSR stimulates glucose uptake. INSR and IGF-1 receptors share major structural and functional similarity. The earliest cellular response to insulin stimulation is autophosphorylation of tyrosine in INSR. In humans, the INSR gene is located on chromosome 19. Defects in INSR are the cause of various insulin resistance syndromes and IGF-1R defects may also cause some forms of growth retardation.
