Immunofluorescence analysis of ISL1 was performed using 70% confluent log phase SH-SY5Y cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. ISL1 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of ISL1 was performed using 70% confluent log phase SH-SY5Y cells. The cells were labeled with TCF7 Monoclonal Antibody (3D3) (Product # MA5-32206) at 1:200 dilution in 0.1% BSA, incubated at 4 degree Celsius overnight and then labeled with Donkey anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor Plus 488 (Product # A32790) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing localization to nucleus. Panel e shows HeLa cells with no expression of ISL1. Panel f represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

ISL1 Antibody in Immunocytochemistry (ICC/IF) — Immunocytochemical analysis of ISL1 in A549 cells using a ISL1 Monoclonal antibody (Product # MA5-32206) as seen in green. The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

ISL1 Antibody in Western Blot (WB) — Western blot was performed using ISL1 Recombinant Rabbit Monoclonal Antibody (SC05-64) (Product # MA5-32206) and a ~42 kDa band corresponding to ISL1 was observed in SHSY5Y; and was low or absent in SKOV3, HeLa and THP1. Whole cell lysate (30 µg lysate) of SHSY5Y (Lane 1), SKOV3 (Lane 2), HeLa (Lane 3) and THP1 (Lane 4) were electrophoresed using NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

ISL1 Antibody in Western Blot (WB) — Western blot analysis of ISL1 in MCF-7 cell lysate using a ISL1 Monoclonal antibody (Product # MA5-32206) at a dilution of 1:1,000.

ISL1 Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across tissues tested owing to their inherent genetic constitution. Relative expression of ISL1 was observed in SHSY5Y in comparison to SKOV3, HeLa and THP1 using ISL1 Recombinant Rabbit Monoclonal Antibody (SC05-64) (Product # MA5-32206) in Western Blot. {RE}

Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The expression systems are developed by cloning in the specific antibody DNA sequences from immunoreactive rabbits. Then, individual clones are screened to select the best candidates for production. The advantages of using recombinant rabbit monoclonal antibodies include: better specificity and sensitivity, lot-to-lot consistency, animal origin-free formulations, and broader immunoreactivity to diverse targets due to larger rabbit immune repertoire.

This gene encodes a member of the LIM/homeodomain family of transcription factors. The encoded protein binds to the enhancer region of the insulin gene, among others, and may play an important role in regulating insulin gene expression. The encoded protein is central to the development of pancreatic cell lineages and may also be required for motor neuron generation. Mutations in this gene have been associated with maturity-onset diabetes of the young.
