Western blot analysis was performed on nuclear enriched extracts (30 µg lysate) of U-937 (Lane 1).The blots were probed with MBD1 Rabbit Polyclonal Antibody (Product # PA1-889, 2 µg/mL) and detected by chemiluminescence using Goat anti-Rabbit IgG (Heavy Chain) Superclonal Secondary Antibody, HRP conjugate (Product # A27036, 0.4 µg/mL, 1:2500 dilution). PA1-889 detects human methyl CpG binding domain protein 1 (MBD1) recombinant protein.
PA1-889 has been successfully used in Western blot procedures. By Western blot, this antibody detects an ~83 kDa protein representing recombinant human MBD1.

Methyl-CpG-binding domain protein 1 (MBD1) is a member of a family of mammalian proteins that specifically bind methylated regions of DNA. It is a transcriptional repressor that binds CpG islands in promoters where the DNA is methylated at position 5 of cytosine within CpG dinucleotides. Binding is abolished by the presence of 7-mG that is produced by DNA damage by methylmethanesulfonate (MMS). MBD1 acts as a transcriptional repressor and plays a role in gene silencing by recruiting AFT7IP, which in turn recruits factors such as the histone methyltransferase SETDB1. Isoform 1 and isoform 2 can also repress transcription from unmethylated promoters.
