MPO Antibody (DOM00001G-500UG) in ICC/IF
Isolated monocytes were stained with Lysotrack red followed by staining with Rabbit antibody to MPO (3 µg/mL) for 1 hour at room temperature, washed and followed by staining with goat anti-rabbit antibody conjugated to Alexa 488 (Green) for 1 hr. Mounting media (10% Glycerol in PBS) containing Hoechst 33258 1 µg/mL (Sigma-Aldrich) was used for nuclear counterstaining (Blue). Fluorescent cell staining was analyzed by confocal microscopy using a Bio-Rad Radiance 2100 confocal microscope. Magnification: 200X.

Myeloperoxidase (MPO) is a heme protein that is synthesized during myeloid differentiation and is a major component of neutrophil azurophilic granules. It is produced as a single chain precursor and subsequently cleaved into a light and heavy chain. The mature MPO is a tetramer composed of 2 light chains and 2 heavy chains. This enzyme is responsible for producing hypohalous acids that are central to the microbicidal activity of neutrophils.
