Myogenin Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of Myogenin in NIH/3T3 cells. Samples were incubated with Myogenin Polyclonal antibody (Product # PA5-116750) using a dilution of 1:100. Blue: DAPI for nuclear staining.

Myogenin Antibody in Western Blot (WB) — Western blot analysis of Myogenin in various lysates. Samples were incubated with Myogenin Polyclonal antibody (Product # PA5-116750) using a dilution of 1:2,000, followed by HRP Goat Anti-Rabbit IgG (H+L) at a dilution of 1:10,000. Lysates/proteins: 25 µg per lane. Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Basic Kit. Exposure time: 90s.

Myogenin Antibody in Western Blot (WB) — Western blot was performed using Myogenin Polyclonal Antibody (Product # PA5-116750) and a 30 kDa band corresponding to MYOG was observed. Nuclear enriched extracts (30 µg lysate) of C2C12 (Lane 1), C2C12 differentiated to myotubes (2% heat inactivated horse serum, 5 days) (Lane 2), RD (Lane 3), MCF7 (Lane 4) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0302BOX), 12 well. Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:20,000 dilution) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580).

Myogenin Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across cell lines owing to their inherent genetic constitution. Relative expression of MYOG was observed in C2C12 differentiated to myotubes and not in undifferentiated C2C12 using Myogenin Polyclonal Antibody (Product # PA5-116750) in Western Blot. In addition to this, the expression was observed in RD cells (positive), and not in MCF7 cells (negative). {RE}
