Myogenin Antibody (PA5-85111) in WB

Western blot was performed using Myogenin Polyclonal Antibody (Product # PA5-85111) and a 32 kDa band corresponding to Myogenin was observed along with uncharacterized bands (*) at ~38 kDa and ~150 kDa. Myogenin Antibody in Western Blot (WB) — Western blot was performed using Myogenin Polyclonal Antibody (Product # PA5-85111) and a 32 kDa band corresponding to Myogenin was observed along with uncharacterized bands (*) at ~38 kDa and ~150 kDa. Whole cell extracts (30 µg lysate) of RD (Lane 1) and HEK-293 (Lane 2) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0302BOX), 12 well. Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB33001) by iBlot™ 3 Western Blot Transfer Device (Product # IB31001). The blot was probed with the primary antibody (1:1,000) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:20,000) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580). Band of interest was not observed in HEK-293 which does not express Myogenin.

Myogenin Antibody in Western Blot (WB) — Knockdown of Myogenin was achieved by transfecting RD with Myogenin specific siRNAs (Silencer® select Product # s9232,s9233). Western blot analysis (Fig. a) was performed using Whole cell extracts from the Myogenin knockdown cells (lane 3), non-targeting scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with Myogenin Polyclonal Antibody (Product # PA5-85111, 1:1,000) and Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:20,000). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to Myogenin.

Myogenin Antibody in Western Blot (WB) — Western blot analysis of Myogenin in whole cell extracts using Myogenin polyclonal antibody (Product # PA5-85111) using 30 µg of sample at a dilution of 1:500. Prior to incubation with primary antibody, the sample was separated on 12% SDS-PAGE.

Myogenin is a muscle-specific transcription factor that can induce myogenesis in a variety of cell types in tissue culture. It is a member of a large family of proteins related by sequence homology, the helix-loop-helix proteins. It is essential for the development of functional skeletal muscle.
