NUMB Antibody (A301-715A) in IP

Detection of human NUMB by western blot of immunoprecipitates. Samples: Whole cell lysate (1 mg for IP, 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-NUMB antibody (Product # A301-715A) used for IP at 3 µg/mg lysate. NUMB was also immunoprecipitated by rabbit anti-NUMB antibody A301-714A, which recognizes an upstream epitope. For blotting immunoprecipitated NUMB, A301-714A was used at 0.1 µg/mL. Detection: Chemiluminescence with an exposure time of 30 seconds.

Protein NUMB homolog is an evolutionarily conserved protein that controls the differentiation of neuronal progenitor cells. Recent studies have revealed that NUMB functions in clathrin-dependent endocytosis by binding to the AP-2 complex. NUMB is also expressed in postmitotic neurons and plays a role in axonal growth. Neural cells expressing Numb isoforms with short phosphotyrosine-binding (SPTB) domain undergo extensive neurite outgrowth, an effect that can be blocked by voltage-gated Ca2+ channel (VGCC) inhibitor or by Ca2+ chelator.
