Ozuriftamab is a biosimilar targeting the receptor tyrosine kinase orphan receptor 2 (ROR2). The ROR2 gene encodes the protein receptor tyrosine kinase-like orphan receptor 2 (ROR2), which is involved in non-canonical Wnt signaling pathways. ROR2 is known to play critical roles in skeletal development, particularly in the formation of limb bones and cartilage. The gene is located on chromosome 9 and consists of 9 exons that encode a protein of 943 amino acids with a single-pass transmembrane receptor structure. ROR2's extracellular region contains an immunoglobulin-like domain, a cysteine-rich domain (CRD), and a kringle domain, which facilitate its interaction with Wnt ligands such as Wnt5a and Wnt11. Upon ligand binding, ROR2 can homodimerize to initiate downstream signaling cascades that influence cell polarity, migration, and differentiation. Aberrations in ROR2 expression and function are implicated in various disorders, including Robinow syndrome and recessive renal cancer. Additionally, ROR2 has been noted to play a role in cancer cell motility and invasion, highlighting its potential as a therapeutic target.
