PLCG1 Antibody (PA5-99510) in ICC/IF

Immunocytochemistry analysis of PLCG1 in Hela cells. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100, then blocked in 10% serum for 45 minutes at 25°C. Samples were incubated with PLCG1 polyclonal antibody (Product # PA5-99510) and mouse anti-beta tubulin antibody for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) (Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) (Green) were used as the secondary antibodies. The nuclear counter stain is DAPI (blue).

Antibody detects endogenous levels of total PLCG1.

The mammalian Phospholipase Cg (PLCg) family has two closely related proteins, PLCg1 and PLCg2. The PLCg isozymes have the core structure domains and a unique array of domains containing an additional PH domain, two SH2 domains and one SH3 domain. In response to extracellular stimuli, such as hormones and growth factors, receptor tyrosine kinases (RTKs) phosphorylate and activate PLCg. Activated PLCg catalyzes hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) to produce the metabolic second messenger molecules inositol 1,4,5-triphosphate (IP3) and diacylglycerol (DAG).
