Immunofluorescence analysis of RSK1 was performed using 70% confluent log phase NTERA-2 cl.D1 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.01% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. RPS6KA1 (RSK1) is a growth-factor regulated serine/threonine kinase that is involved in the MAPK cascade. It is thought to control cellular proliferation and differentiation through the phosphorylation of transcription factors. RPS6KA1 contains 2 nonidentical kinase catalytic domains and phosphorylates various substrates, including members of the mitogen-activated kinase (MAPK) signalling pathway. The activity of this protein has been implicated in controlling cell growth and differentiation. Alternate transcriptional splice variants, encoding different isoforms, have been characterized.
