Immunofluorescence analysis of SFRP1 was performed using 70% confluent log phase MCF 10A cells treated with PTI (1X for 4hrs). SFRP1 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of SFRP1 was performed using 70% confluent log phase MCF 10A cells treated with PTI (1X for 4hrs). The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. The cells were labeled with SFRP1 Recombinant Rabbit Monoclonal Antibody (24) (Product # MA5-30873) at 1:100 dilution in 0.1% BSA, incubated at 4 degree Celsius overnight and then labeled with Donkey anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor Plus 488 (Product # A32790, 1:2000 dilution), for 45 minutes at room temperature (Panel a, e: Green). Nuclei (Panel b, f: Blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c, g: Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300 dilution). Panel h represents the merged image showing cytoplasmic as well as Golgi like localization in MCF 10A cells treated with PTI. Panel (a-d) represent MCF 10A cells. Panel (e-h) represents MCF 10A cells treated with PTI (1X for 4hrs). The images were captured at 60X magnification. Increased accumulation of SFRP1 was seen in Golgi with PTI treatment.

SFRP1 Antibody in Western Blot (WB) — Western blot was performed using Anti-SFRP1 Recombinant Rabbit Monoclonal Antibody (24) (Product # MA5-30873) and a 40 kDa band corresponding to SFRP1 was observed across cell lines tested. Whole cell extracts (30 µg lysate) of A549 (Lane 1), A549 treated with PTI (1X for 4hrs) (Lane 2), MCF 10A (Lane 3), MCF 10A treated with PTI (1X for 4hrs) (Lane 4), SK-BR-3 (Lane 5) and SK-BR-3 treated with PTI (1X for 4hrs) (Lane 6) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX), 12 well. Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:20,000) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580).Expression of SFRP1 was observed to be high in untreated as well as treated cells of A549 and MCF 10A in comparison to SK-BR-3.

SFRP1 Antibody in Western Blot (WB) — Knockdown of SFRP1 was achieved by transfecting MCF 10A with SFRP1 specific siRNAs (Silencer® select Product # S12714, S12715). Western blot analysis (Fig. a) was performed using Whole cell extracts from the SFRP1 knockdown cells (lane 3), non-targeting scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with SFRP1 Recombinant Rabbit Monoclonal Antibody (24) (Product # MA5-30873, 1:1000 dilution) and Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:20,000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to SFRP1.

Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. Secreted frizzled-related protein 1 (SFRP1) is a member of the SFRP family that contains a cysteine-rich domain homologous to the putative Wnt-binding site of Frizzled proteins. SFRPs act as soluble modulators of Wnt signaling. SFRP1 and SFRP5 may be involved in determining the polarity of photoreceptor cells in the retina. SFRP1 is expressed in several human tissues, with the highest levels in heart.
