Thrombopoietin Antibody (PA5-95945) in WB
Western blot analysis of Thrombopoietin in extracts of various cell lines. Samples were incubated with Thrombopoietin Polyclonal antibody (Product # PA5-95945) using a dilution of 1:1,000, followed by HRP Goat Anti-Rabbit IgG (H+L) at a dilution of 1:10,000. Lysates/proteins: 25 µg per lane. Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Basic Kit. Exposure time: 180s.

Thrombopoietin (TPO, THPO, MPL ligand, megakaryocyte colony-stimulating factor) is responsible for the generation of platelets through its action on hematopoietic cells committed to the megakaryocyte lineage. Binding of thrombopoietin to its receptor, MPL (CD110), initiates a signaling cascade via JAK/STAT, MAPK, and PI3K/AKT signaling pathways that leads to the proliferation and differentiation of megakaryocytes and, ultimately, to the production of platelets (thrombopoiesis). Thrombopoietin is constitutively produced by the liver and has a predicted molecular weight of 38 kDa.
