NK-92, HeLa, Jurkat, and A549 cells were either untreated or IFN-b-treated for 48 hours as indicated. 8220Transfected8221 (last lane) represents extract from HEK293T cells transfected with plasmids expressing 3X-FLAG-ISG15, UBE1L, UBCH8, and HERC5. UBE1L (ubiquitin-like modifier-activating enzyme 7) activates ubiquitin by first adenylating with ATP at its C-terminal glycine residue and then linking that residue to the side chain of a cysteine residue in E1. This activation results in an ubiquitin-E1 thioester and free AMP. UBE1L also catalyzes the ISGylation of influenza A virus NS1 protein.
