For larger sizes, the number of freeze/thaws may be reduced by preparing aliquots, aliquots below 20 µL are not recommended. Please never store a kinase diluted. This recombinant fusion protein consists of full-length activin A receptor kinase and a glutathione S-transferase (GST) tag. It is validated in the LanthaScreen™ Eu Kinase Binding Assay. No special measures were taken to activate this kinase.

Easily Screen for Kinase Binding
The LanthaScreen™ Eu Kinase Binding Assay is a TR-FRET assay that lets you measure inhibitor binding to non-activated kinases, perform simple off-rate measurements, and makes simple the study of type III (allosteric) inhibitors.
The LanthaScreen™ Eu Kinase Binding Assay is easy to optimize and run. The assay uses a simple mix-and-read format with a single one hour incubation and no development steps. Simultaneous binding of an Alexa Fluor™ conjugate or “tracer” to a tagged kinase is detected by addition of a europium-labeled anti-tag antibody, in this case anti-GST. Simultaneous binding of the tracer and antibody to the kinase results in a high degree of fluorescence resonance energy transfer (FRET), while displacement of the tracer with a kinase inhibitor results in a loss of FRET (Fig 1).

Protein Form: Full Length
Activin A receptor kinase (ACVR2A) is a type II member of the TGF-beta family of receptor Serine/Threonine kinases. ACVR2A is involved in the activin and myostatin signaling pathway. (ACVR2A Aliases: ACVR2, ACTRII). These receptors are all transmembrane proteins, composed of a ligand-binding extracellular domain with cysteine-rich region, a transmembrane domain, and a cytoplasmic domain with predicted serine/threonine specificity. Type I receptors are essential for signaling, and type II receptors are required for binding ligands and for expression of type I receptors. Type I and II receptors form a stable complex after ligand binding, resulting in phosphorylation of type I receptors by type II receptors. Type II receptors are considered to be constitutively active kinases.
